Descripción : | The aim of the present study was to evaluate the antioxidant effect of a new steroid (4-chloro-17α-acetoxy-4-pregnene-3,20-dione) in rat brain. Twelve adults rats of 500g were randomly distributed in groups of 6 that were treated with 4-chloro-17α-acetoxy-4-pregnene-3,20-dione (4 mg/kg/day), or vehicle, administered during 5 days. At the end of treatment all animals were sacrificed and the GSH levels in the brain were measured by fluorescence method, as well as lipid peroxidation (TBARS), Na+,K+-ATPase and total ATPase enzymatic activity by spectrophotometry methods. Na +,K+-ATPase and total ATPase activities increased, only the total ATPase activity increase was significant as compared to the control group (p<0.05). In the group that received 4-chloro-17α-acetoxy-4- pregnene-3,20-dione the GSH levels remained equal, as compared to the control group. The most important alteration was observed in the lipid peroxidation levels of the steroid group, because TBARS levels diminished significantly (p<0.05) in the group receiving the steroid vs the control group. The results of this study suggest that the steroid 4-chloro-17α-acetoxy-4-pregnene-3, 20-dione in a single dose in short period of treatment altered the lipid peroxidation and the membrane fluidity both in brain adult animals, as consequence of their structural changes. ©INNN, 2007. |
